Research background
PNC-27 is a 32-amino-acid synthetic peptide made of two parts: residues 12-26 of p53 (PPLSQETFSDLWKLL), which form the domain through which p53 binds its regulator HDM-2, and a seventeen-amino-acid segment of the Antennapedia protein, described as a membrane-penetrating sequence.
The reference work is laboratory-based (Sarafraz-Yazdi et al., PNAS 2010). The authors describe HDM-2 in the membranes of several tumour cell lines but not in those of several untransformed lines, and observe that the peptide binds this membrane HDM-2 and causes cell lysis. Other work studies it through theoretical models of its structure. None of the PubMed-indexed articles on PNC-27 is classified as a clinical trial.
Reconstitution and handling
- Work on a clean surface and disinfect the vial stopper with an alcohol wipe before piercing it.
- Add the bacteriostatic water slowly, letting it run down the inner wall of the vial rather than landing on the powder.
- Do not shake. Swirl the vial gently until the lyophilized powder has fully dissolved: vigorous agitation degrades the peptide.
- The solution should come out clear. If it turns cloudy or shows suspended particles, do not use it.
Storage and stability
- Before reconstitution: refrigerated between 2 and 8 °C, protected from light and moisture.
- After reconstitution: in the fridge and away from light. In solution the peptide lasts far less than as a powder, so it is best used soon.
- Avoid freeze-thaw cycles.
