Research background
AOD-9604 reproduces the C-terminal region of human growth hormone that researchers at Monash University identified as its lipolytic domain. It is a sixteen-amino-acid peptide with a disulfide bridge: fragment 177-191 plus a tyrosine at the N-terminus.
The reference work is in rodents. In obese Zucker rats, the authors describe increased lipolytic activity in adipose tissue and, unlike the intact hormone, no adverse effect on insulin sensitivity measured by euglycaemic clamp (Ng et al., Horm Res 2000). In obese mice, increased expression of the beta-3 adrenergic receptor was observed, and the effect on body weight did not appear in mice lacking that receptor (Heffernan et al., Endocrinology 2001). It also features in the detection methods of anti-doping laboratories.
Reconstitution and handling
- Work on a clean surface and disinfect the vial stopper with an alcohol wipe before piercing it.
- Add the bacteriostatic water slowly, letting it run down the inner wall of the vial rather than landing on the powder.
- Do not shake. Swirl the vial gently until the lyophilized powder has fully dissolved: vigorous agitation degrades the peptide.
- The solution should come out clear. If it turns cloudy or shows suspended particles, do not use it.
Storage and stability
- Before reconstitution: refrigerated between 2 and 8 °C, protected from light and moisture.
- After reconstitution: in the fridge and away from light. In solution the peptide lasts far less than as a powder, so it is best used soon.
- Avoid freeze-thaw cycles.
