Research background

DSIP was born of one specific experiment: Monnier and Schoenenberger induced sleep by electrical stimulation in rabbits, collected cerebral venous blood and isolated from it a nonapeptide which, transferred to other animals, was associated with an increase in delta waves. Hence the name, which describes the assay it appeared in and not a mechanism.

Half a century later that mechanism is still not settled, and that is what makes DSIP interesting as a tool: no receptor of its own has been identified for it. The literature studies it by indirect routes — modulation of ACTH and LH release, thermoregulation, interaction with opioidergic systems — and still debates whether it circulates as an endogenous peptide with a function of its own or as a fragment of something larger.

It is also one of the few peptides of its size described as crossing the blood-brain barrier in both directions, and that property has made it a recurring reference in work on the transport and stability of small peptides.

Reconstitution and handling

  • Work on a clean surface and disinfect the vial stopper with an alcohol wipe before piercing it.
  • Add the bacteriostatic water slowly, letting it run down the inner wall of the vial rather than landing on the powder.
  • Do not shake. Swirl the vial gently until the lyophilized powder has fully dissolved: vigorous agitation degrades the peptide.
  • The solution should come out clear. If it turns cloudy or shows suspended particles, do not use it.

Storage and stability

  • Before reconstitution: refrigerated between 2 and 8 °C, protected from light and moisture.
  • After reconstitution: in the fridge and away from light. In solution the peptide lasts far less than as a powder, so it is best used soon.
  • Avoid freeze-thaw cycles.
