Research background
Natural GHRH loses its activity within minutes: DPP-4 cleaves off its first two amino acids. The 1-29 fragment retains the full activity of the whole hormone, and onto it four substitutions were introduced — D-Ala at 2, Gln at 8, Ala at 15, Leu at 27 — that slow down that degradation and aggregation. The result is this molecule, with a plasma half-life of about 30 minutes, producing discrete GH pulses rather than a sustained elevation.
The name is worth looking at closely, because it is the most widespread confusion in this group: 'CJC-1295 without DAC' and 'CJC-1295 with DAC' are not the same molecule with a different duration, they are two distinct compounds — C152H252N44O42 and 3367.90 Da for this one; C165H269N47O46 and 3647.25 Da for the other — and the literature itself has equated them by mistake more than once. This is the one that preserves pulsatility, and the one chosen when the experimental design needs it intact.
Reconstitution and handling
- Work on a clean surface and disinfect the vial stopper with an alcohol wipe before piercing it.
- Add the bacteriostatic water slowly, letting it run down the inner wall of the vial rather than landing on the powder.
- Do not shake. Swirl the vial gently until the lyophilized powder has fully dissolved: vigorous agitation degrades the peptide.
- The solution should come out clear. If it turns cloudy or shows suspended particles, do not use it.
Storage and stability
- Before reconstitution: refrigerated between 2 and 8 °C, protected from light and moisture.
- After reconstitution: in the fridge and away from light. In solution the peptide lasts far less than as a powder, so it is best used soon.
- Avoid freeze-thaw cycles.
